 note = {【目的】ウシ胎児血清（Fetal Bovine Serum：FBS）は、脂肪細胞に分化する性質を有するマウス線維芽細胞3T3-L1を培養する上で良く利用されている。本研究は、汎用性とコスト面を考慮したFBSに代わる手段を提案することを目的とし、FBS代替品血清が3T3-L1に及ぼす影響を調べた。
【方法】FBS代替品血清EquaFETALを使用して3T3-L1の分化誘導を行い、脂肪滴の蓄積量とアディポサイトカインの一種であるアディポネクチン分泌量を経時的に測定した。さらに、分化誘導後Day８とDay14において、アディポネクチン分泌が誘導される条件を検討した。
【結果】3T3-L1の培養にFBS代替品血清を使用することで、脂肪滴の蓄積能を有する脂肪細胞に分化誘導可能であることが示された。アディポネクチンの基礎分泌量に対しては、FBS代替品血清が分化誘導開始後Day８～10をピークに、FBS血清培養条件と比べて早い段階から減少させる可能性が示唆された。アディポネクチン分泌誘導因子の効果は、今回の実験条件では確認できなかった。
【結論】本研究で得られた知見は、3T3-L1をFBS代替品血清で分化誘導した報告が少ない現状において、意義のある基礎的データとなりうると考えられる。しかしながら、アディポネクチン分泌が誘導および維持される条件は、今後さらに詳細な検討を行う必要がある。, Fetal bovine serum (FBS) has widely been used for growth and maintenance of cell cultures. 3T3-L1 is one of the most commonly used preadipose cell lines and is characterized by its differentiation from the fibroblastic phenotype to adipocytes. Given its versatility and cost-effectiveness, the FBS alternative “EquaFETAL®” should be available for conventional use. The present study examined the experimental conditions necessary for the sufficient storage of 3T3-L1 lipid droplets and secreted adiponectin using EquaFETAL®. Oil Red O staining showed that EquaFETAL® maintained the ability of 3T3-L1 adipocytes to accumulate lipid droplets. Adiponectin production peaked at days 8 to 10 of the differentiation periods and tended to decrease until day 14. These findings are considered significant as few reports have examined the influence of EquaFETAL® on 3T3-L1 culture. However, adiponectin secretion-inducing factor had little effect on 3T3-L1 culture in the present study.
